What are the most important changes to know?

Finally just when I felt comfortable with the best move sets, attackers, defenders and battle mechanics they change it. So what has changed with the Pokémon I already had? Move sets and rankings? Which new Pokemon should I be focusing on for powering up and adding to my attack and defender teams?

Johnny - Too broad of a (first) question and not enough detail to offer a specific reply, but in general, some, but not all, pokemons got different move sets. Some got better, some got worse, some just got different. Battle mechanics did not change. I do not have answers to your other questions.

Was trying to get just a few people's opinion on the major changes they have found that affected there stable of mons the most? Was just starting to go through my attack teams and was trying to see which ones I need to replace cause either there is a new one better or have to evolve another of the same cause their move set in now inferior.

Example is confusion now the better then psycho cut ? I know Tyranitar is the new top CP attacker but is there any other Gen 2 mons that out perform the Gen 1 mons? Blissey I have been hearing is now a top if not the best defender. This true or what? Any other Gen 2 defenders should I be working on leveling?

Confusion hits harder but slower than Pc. Sure it is more dps for you, but it is also more dps against you as the dodge is clunky.

Blissey is by far the new 'best' defender.

You now have a choice with nearly all mons - do you want to stand still and beat on the opponent with big dps, or do you want to dance around and dodge everything and build fast energy for a big KO...or dance with the enemy, and wittle away at their stamina with a series of smaller damaging hits from multi bar charge moves
You can now choose.

I have issues with lag often so I am just happy when I can dodge the charge moves. So my attack team is tanky. Mostly use Nites against tanky defenders , Laprasses against Nites and Rhydons and Evee evolutions to type advantages against the rest. Hope this helps with my questions?

The biggest change is that most moves are useful. Some are better than others, some got nerfed, others got buffed; but few are useless.

I basically evolved a Blissey (P/DG 91%iv) and powered it up to 2797 now and put it into a good gym. And switched buddy from Snor to Chansey. And powering up C/SoBs to kick other one's Blisseys. Other than that, not enough stardust to do anything!

Some changes that hit existing pokemon hard:
- Psychic and Thunder are much worse than they were before, making pokemon with them (notably Alakazam and Jolteon) much worse.
- Wigglytuff is no longer as useful as an all-around prestiger.

New pokemon you'll want to pursue:
- Machamp with Counter/Dynamic Punch is an elite attacker.
- Tyranitar supplants Dragonite as a gym topper.
- Blissey is in a tier of her own as a gym defender.

This is a great response, I think you hit all the major points.

Why do you say wigglytuff is less useful as an all-around prestiger? The only case I can think of is wigglytuff doesn't work that well when prestiging a blissey, because she tends to run out of time. Other than that, she seems to me to work as well as before.

Fully agree!

It works even better than before, because Pound + any of her charged attacks works great now.

The biggest change is that quick moves are now balanced. The moves with the highest DPS have the lowest EPS and the moves with the best EPS have the worst DPS. Charge moves are also now much stronger. Unlike gen 1 where DPS was a critical stat with Gen 2 moves its about attacks per 100 energy and damage per move, DPS is deceiving imo.
As far as whats the best mons and movesets its too early to say. However any serious trainer needs an army of Machamps.
